The land Framingham sits on was initially inhabited by the Nipmuck tribe and is located on what was an important Native American trail that led from MA Bay to the Connecticut River Valley. The first settler, John Stone, arrived in 1647. Others followed, and a couple of decades later, 15,000 acres were acquired by Thomas Danforth, and Framingham was incorporated in 1700.

From 1854 to 1865, it was a regular gathering spot for activists in the abolitionist movement where members of the Massachusetts Anti-Slavery Society would meet, including members such as Henry David Thoreau and William Garrison. In 1948, Framingham embarked on a healthcare research project known as the Framingham Heart Study, which sought to find common causes of cardiovascular disease and improve the population's health.
